A study on the effect of Tamoxifen (Nolvadex) on the hypothalamic-pituitary-gonadal axis was carried out by Li et al. (2020) in a group of men with elevated LH and/or testosterone levels and a small number of subjects with normal sperm parameters (1). The aim of the study was to examine whether the effect of Nolvadex (Tamoxifen) on testosterone levels could be reversed by administering it to healthy volunteers and whether it could also be used to treat testosterone deficiency. The study was conducted in two phases. The first phase of the study was a crossover study in which the effect of Tamoxifen (Nolvadex) was compared with that of Tamoxifen alone and in combination with testosterone (Tamoxifen + Tamid). Tamoxifen was administered to healthy subjects (n=12), who were diagnosed with normal testosterone levels. The second phase of the study was an open-label, controlled study in which subjects were randomly allocated to two groups (Tamoxifen + Nolvadex and Tamoxifen + Tamid). Subjects were assigned to receive Tamoxifen (Nolvadex) and Tamoxifen + Tamid (Tamoxifen + Nolvadex) for 3 years. The study was conducted in both groups on the basis of a randomized, double-blind, placebo-controlled study design. In the Tamoxifen group, there were significant decreases in LH and/or testosterone levels compared with the control group, which were similar to those observed in men who received Tamoxifen alone, as well as to those in men who received Tamoxifen alone and Tamoxifen + Tamid. In the Tamoxifen + Tamid group, there was also an increase in LH levels compared with the Tamoxifen + Tamid group. The effect of Tamoxifen + Tamid was comparable to that of Tamoxifen alone. The results of the study were comparable to those observed in men who received Tamoxifen alone and in men who received Tamoxifen + Tamid. The results were also comparable to those observed in men who received Tamoxifen + Tamid. These results showed that Nolvadex (Tamoxifen) was no more effective than Tamoxifen alone in treating testosterone deficiency, but Nolvadex was more effective than Tamoxifen alone in treating LH and/or testosterone deficiency. However, the overall result was the opposite: the effect of Tamoxifen was similar to that of Tamoxifen alone. In the presence of elevated testosterone levels, Nolvadex should be considered a suitable treatment in men who are predisposed to the adverse effect of testosterone deficiency (2,3).

Side effects of NOLVADEX: Some of the common side effects of NOLVADEX are nausea, diarrhea, hot flushes, restlessness, pain in the back, breasts, mood, hair and bone reactions, and skin rash.
